Using gas chromatography–mass spectrometry (GC–MS) with both electron-impact and chemical-ionization modes on polar and non-polar columns, the researchers characterized the volatile components of the essential oil extracted from Cyperus rotundus tubers. The analysis revealed over 33 constituents, with cyperene, ?-cyperone, isolongifolen-5-one, rotundene, and cyperorotundene being the dominant compounds, together accounting for about 62% of the total oil.
MTT-based cytotoxicity testing showed that the essential oil exhibited strong inhibitory effects on the L1210 leukemia cell line, which was further supported by a marked increase in apoptotic DNA fragmentation. To assess its antioxidant potential, the oil was examined through several assays, including DPPH radical scavenging, the xanthine/xanthine oxidase system, and superoxide scavenging generated through riboflavin photo-reduction. Its antimutagenic capacity was evaluated by monitoring the reduction of DNA strand breaks in a pBS plasmid caused by H?O? under UV exposure.
